If you're running a medium to large pedalboard, then a buffer is definitely recommend. While True Bypass pedals are great, you also need something to drive the signal through the chain. Even using high quality cables in a large rig will never sound as good as 5ft of average cable plugged directly into an amp.

Put it first in the chain of effects, plug in your input and output cables and a 9V power supply (like the 1 SPOT...), and your guitar will sound like it's plugged directly into your amp with a short cord.
